Concentration Determination

In manufacturing biologics, it is crucial to understand the level of protein produced at various times. Label-free binding analysis with the SKi Pro™ platform offers a way of performing these measurements without the need to purify the analyte mixture.

An antibody molecule, designed to interact with the biologic analyte, is immobilized onto the SKi Sensor™ multiwell strips. These strips are then dipped and read, eight wells at a time, into a 96-well plate containing unpurified aliquots from the biological mixture. The rate at which analytes bind to their antibodies on the surface is porportional to the concentration of the analyte in the mixture. So by measuring this rate, one can determine the actual concentration by use of a calibration curve.

For antibodies which can be regenerated, a single SKi Sensor™ well strip is often sufficient to characterize an entire 96-well plate, which keeps consumable costs for the assay well below $1/well.
